The Dance For Life "dance-a-thon parties" will be individual three-hour events taking place over seven nights at seven arenas across the UK.
Each will see the public take to the floor, with Kay acting as "DJ PK" on the turntables.
Kay said: "I can't think of a more enjoyable way to raise money for such an important charity."
He added that he was hoping for a lot of public support and that those taking part would get the chance to dance "to some of the best music of the last four decades".
Simon Ledsham from Cancer Research UK, said: "We are thrilled that Peter is holding these dance-a-thons for the charity.
"We rely on the donations of supporters to fund vital research to help us beat cancer sooner, so it's brilliant to have his support and we hope the public all get their dancing shoes on and join him."
The dance marathons will start in January and run until April and will span the length and breadth of the UK, in cities including Liverpool, Cardiff, Aberdeen and Belfast.
Each arena will be transformed to accommodate vast dance floors with "state of the art" sound and lights.
All the profits from the ticket sales for Dance For Life will go towards Cancer Research UK
